Pre-transmuxing MP4

Currently, I used hls vod streaming

If there are many connection, internal server error will be occurs.
Can I upload .ts files to the server instead of .mp4 then making nimble streamer serve already transmuxing content ?

An "internal server error" is a point to investigate first;
1. Take a look at /var/log/nimble/nimble.log for any related error messages " E: ", perhaps some fine tuning is needed;
2. Take a look at CPU load per-thread.
If any thread of Nimble is loaded - a fine tuning is applicable according to the machine's capabilities:

I'd suggest opening a Support ticket here: https://wmspanel.com/help , please provide more detailed information in the ticket;
Distributing a pre-transmuxed content is also possible, you can create a VOD route to a directory, where you can place a 'playlist.m3u8' with 'chunk.m3u8' with all the (.ts) chunks.
