How to run multiple instances of Nimble Streamer on one computer?

It's not available but why would you need to run several instances?

Assume:
1. A one instance for dvr functionality only.
2. A second one as VOD gateway.
3. 3rd LIVE gateway.
4. ...
In another words instance per "feature".

Single Nimble instance can handle all this.
You may want to add more worker threads in config file http://blog.wmspanel.com/p/nimble-streamer-configuration.html
But this will be required only in case of heavy duty.

Are different machines?

Yes, of couse. These are different servers of your delivery network.
